The church already existed in the Middle Ages with the name of St. Mary del Masaccio. It was rebuilt at the end of the 15th century and named after St. Roch.
Papers state the main proponent and supporter of this new Renaissance building were Francesco di Pietrasanta. The church features a Greek cross plant and was never finished outside.
